Uncertain significance for Breast-ovarian cancer, familial, susceptibility to, 1 — the classification assigned by KCCC/NGS Laboratory, Kuwait Cancer Control Center to NM_007294.4(BRCA1):c.1907G>T (p.Cys636Phe), citing ACMG Guidelines, 2015. This variant lies in the BRCA1 gene (transcript NM_007294.4) at coding-DNA position 1907, where G is replaced by T; at the protein level this means replaces cysteine at residue 636 with phenylalanine — a missense variant. Submitter rationale: This sequence change replaces cysteine, which is neutral and slightly polar, with phenylalanine, which is neutral and non-polar, at codon 636 of the BRCA1 protein (p.Cys636Phe).This amino acid position is not well conserved (PhyloP=-013). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with BRCA1-related conditions. In addition, this alteration is predicted to be tolerated by in silico analysis. In summary, the available evidence is currently insufficient to determine the role of this variant in disease.
