Uncertain significance for Colorectal cancer, hereditary nonpolyposis, type 7 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001040108.2(MLH3):c.2726G>C (p.Ser909Thr), citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (gnomAD no frequency). This sequence change replaces serine, which is neutral and polar, with threonine, which is neutral and polar, at codon 909 of the MLH3 protein (p.Ser909Thr). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals affected with MLH3-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated.
