NM_003000.3(SDHB):c.344G>A (p.Arg115Gln) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R115Q variant (also known as c.344G>A), located in coding exon 4 of the SDHB gene, results from a G to A substitution at nucleotide position 344. The arginine at codon 115 is replaced by glutamine, an amino acid with highly similar properties. This variant was identified in a cohort of 681 ancestrally diverse, healthy subjects (Bodian DL et al. PLoS One, 2014 Apr;9:e94554). This amino acid position is well conserved through mammals. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
