NM_004304.5(ALK):c.2416C>A (p.Arg806Ser) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R806S variant (also known as c.2416C>A), located in coding exon 14 of the ALK gene, results from a C to A substitution at nucleotide position 2416. The arginine at codon 806 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
