This variant lies in the NEFL gene (transcript NM_006158.5) at coding-DNA position 26, where A is replaced by G; at the protein level this means replaces tyrosine at residue 9 with cysteine — a missense variant. Submitter rationale: This sequence change replaces tyrosine with cysteine at codon 9 of the NEFL protein (p.Tyr9Cys). The tyrosine residue is highly conserved and there is a large physicochemical difference between tyrosine and cysteine. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with NEFL-related conditions. ClinVar contains an entry for this variant (Variation ID: 1350771).
