Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000455.5(STK11):c.865A>T (p.Met289Leu), citing Ambry Variant Classification Scheme 2023: The p.M289L variant (also known as c.865A>T), located in coding exon 7 of the STK11 gene, results from an A to T substitution at nucleotide position 865. The methionine at codon 289 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
