Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.536A>G (p.Glu179Gly), citing Ambry Variant Classification Scheme 2023: The p.E179G variant (also known as c.536A>G), located in coding exon 6 of the POLE gene, results from an A to G substitution at nucleotide position 536. The glutamic acid at codon 179 is replaced by glycine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
