Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005726.6(TSFM):c.655G>A (p.Gly219Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine, which is neutral and non-polar, with arginine, which is basic and polar, at codon 240 of the TSFM protein (p.Gly240Arg). This variant is present in population databases (rs752963960, gnomAD 0.005%). This variant has not been reported in the literature in individuals affected with TSFM-related conditions. ClinVar contains an entry for this variant (Variation ID: 1348922). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt TSFM protein function with a negative predictive value of 80%.
