Uncertain significance for EGFR-related lung cancer — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005228.5(EGFR):c.1178A>C (p.Asp393Ala), citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (ExAC no frequency). This sequence change replaces aspartic acid with alanine at codon 393 of the EGFR protein (p.Asp393Ala). The aspartic acid residue is weakly conserved and there is a moderate physicochemical difference between aspartic acid and alanine. This variant has not been reported in the literature in individuals with EGFR-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
