Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000302.4(PLOD1):c.1451G>C (p.Cys484Ser), citing Ambry Variant Classification Scheme 2023: The p.C484S variant (also known as c.1451G>C), located in coding exon 13 of the PLOD1 gene, results from a G to C substitution at nucleotide position 1451. The cysteine at codon 484 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
