Uncertain significance for Deficiency of isobutyryl-CoA dehydrogenase — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_014384.3(ACAD8):c.782G>A (p.Ser261Asn), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces serine with asparagine at codon 261 of the ACAD8 protein (p.Ser261Asn). The serine residue is weakly conserved and there is a small physicochemical difference between serine and asparagine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with ACAD8-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt ACAD8 protein function.
