Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001378452.1(ITPR1):c.4912G>T (p.Val1638Phe), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces valine, a(n) neutral and non-polar amino acid, with phenylalanine, a(n) neutral and non-polar amino acid, at codon 1614 of the ITPR1 protein (p.Val1614Phe). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with ITPR1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
