Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000122.2(ERCC3):c.1451T>C (p.Ile484Thr), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the ERCC3 gene (transcript NM_000122.2) at coding-DNA position 1451, where T is replaced by C; at the protein level this means replaces isoleucine at residue 484 with threonine — a missense variant. Submitter rationale: This sequence change replaces isoleucine, which is neutral and non-polar, with threonine, which is neutral and polar, at codon 484 of the ERCC3 protein (p.Ile484Thr). This variant is present in population databases (rs371767613, gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of xeroderma pigmentosum/Cockayne syndrome (Invitae). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 1346717). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
