Uncertain significance for Dilated cardiomyopathy 1R; Hypertrophic cardiomyopathy 11; Atrial septal defect 5 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005159.5(ACTC1):c.410C>T (p.Ala137Val), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the ACTC1 gene (transcript NM_005159.5) at coding-DNA position 410, where C is replaced by T; at the protein level this means replaces alanine at residue 137 with valine — a missense variant. Submitter rationale: This sequence change replaces alanine with valine at codon 137 of the ACTC1 protein (p.Ala137Val). The alanine residue is highly conserved and there is a small physicochemical difference between alanine and valine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with ACTC1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
