Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002460.4(IRF4):c.512T>C (p.Met171Thr), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals with IRF4-related conditions. This variant is present in population databases (rs755905242, ExAC 0.02%). This sequence change replaces methionine with threonine at codon 171 of the IRF4 protein (p.Met171Thr). The methionine residue is moderately conserved and there is a moderate physicochemical difference between methionine and threonine.
