Uncertain significance for Jeune thoracic dystrophy; Nephronophthisis — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_024753.5(TTC21B):c.3102-4C>G, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the TTC21B gene (transcript NM_024753.5) at 4 bases into the intron immediately before coding-DNA position 3102, where C is replaced by G. Submitter rationale: In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. This variant has been observed in individual(s) with nephronophthisis (Invitae). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This variant is present in population databases (rs750756355, gnomAD 0.006%). This sequence change falls in intron 23 of the TTC21B gene. It does not directly change the encoded amino acid sequence of the TTC21B protein.
