Uncertain significance for Primary ciliary dyskinesia 33 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001481.3(DRC4):c.336C>G (p.Asn112Lys), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the DRC4 gene (transcript NM_001481.3) at coding-DNA position 336, where C is replaced by G; at the protein level this means replaces asparagine at residue 112 with lysine — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This sequence change replaces asparagine with lysine at codon 112 of the GAS8 protein (p.Asn112Lys). The asparagine residue is moderately conserved and there is a moderate physicochemical difference between asparagine and lysine. This variant has not been reported in the literature in individuals with GAS8-related conditions. This variant is not present in population databases (ExAC no frequency).
