This variant lies in the GNE gene (transcript NM_005476.7) at coding-DNA position 501, where C is replaced by A; at the protein level this means replaces histidine at residue 167 with glutamine — a missense variant. Submitter rationale: This sequence change replaces histidine, which is basic and polar, with glutamine, which is neutral and polar, at codon 198 of the GNE protein (p.His198Gln). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of autosomal recessive GNE-related conditions (Invitae). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant.
