Uncertain significance for Epidermodysplasia verruciformis — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_152468.5(TMC8):c.1885C>T (p.Arg629Trp), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine with tryptophan at codon 629 of the TMC8 protein (p.Arg629Trp). The arginine residue is moderately conserved and there is a moderate physicochemical difference between arginine and tryptophan. This variant is present in population databases (rs377668539, ExAC 0.01%). This variant has not been reported in the literature in individuals affected with TMC8-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
