NM_004304.5(ALK):c.3744-2A>G was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.3744-2A>G intronic variant results from an A to G substitution two nucleotides upstream from coding exon 25 in the ALK gene. This nucleotide position is highly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice acceptor site and will result in the creation or strengthening of a novel splice acceptor site. However, loss of function of ALK has not been established as a mechanism of disease. Based on the available evidence, the clinical significance of this alteration remains unclear.
