This variant lies in the PRCD gene (transcript NM_001077620.3) at the canonical splice donor site of the intron immediately after coding-DNA position 74, where G is replaced by A; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: Variant summary: PRCD c.74+1G>A alters a conserved nucleotide located in a canonical splice-site and is predicted to affect mRNA splicing resulting in a significantly altered protein due to either exon skipping, shortening, or inclusion of intronic material. Several computational tools predict a significant impact on normal splicing: Three predict the variant abolishes the canonical 5' splicing donor site. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 1.4e-05 in 219476 control chromosomes. c.74+1G>A has been reported in the literature as a homozygous genotype in at-least one individual affected with Retinitis Pigmentosa (example, Perez-Carro_2016).
