Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000030.3(AGXT):c.704C>T (p.Thr235Met), citing LabCorp Variant Classification Summary - May 2015. This variant lies in the AGXT gene (transcript NM_000030.3) at coding-DNA position 704, where C is replaced by T; at the protein level this means replaces threonine at residue 235 with methionine — a missense variant. Submitter rationale: Variant summary: AGXT c.704C>T (p.Thr235Met) results in a non-conservative amino acid change located in the Aminotransferase class V domain (IPR000192) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 1.2e-05 in 251420 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.704C>T in individuals affected with Primary Hyperoxaluria Type 1 and no experimental evidence demonstrating its impact on protein function have been reported. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as uncertain significance.
