Pathogenic for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000219.6(KCNE1):c.20_23dup (p.Val9fs), citing Ambry Variant Classification Scheme 2023: The c.20_23dupCAGC pathogenic mutation, located in coding exon 1 of the KCNE1 gene, results from a duplication of CAGC at nucleotide position 20, causing a translational frameshift with a predicted alternate stop codon (p.V9Sfs*32). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
