This variant lies in the NHS gene (transcript NM_001291867.2) at coding-DNA position 691, where C is replaced by T; at the protein level this means replaces arginine at residue 231 with tryptophan — a missense variant. Submitter rationale: Variant summary: NHS c.691C>T (p.Arg231Trp) results in a non-conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 5.5e-06 in 182500 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance.
