Pathogenic for Severe combined immunodeficiency disease — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NC_000009.11:g.(?_214864)_(399260_404917)del, citing LabCorp Variant Classification Summary - May 2015: Variant summary: The variant identified by MLPA or other technology involves the deletion of exons 1-26 in the DOCK8 gene. A presumed nomenclature of c.(?_-113)_(3234+1_3235-1)del has been designated for the purposes of this classification. Although exact breakpoints of this deletion are not known, it is expected to result in a loss of start in the DOCK8 gene, a known mechanism of disease. The variant was absent in 21640 control chromosomes. c.(?_-113)_(3234+1_3235-1)del has been reported in the literature in individuals affected with primary Immunodeficiency (PLatt_2021). These data indicate that the variant is likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as pathogenic.
