This variant lies in the TEX15 gene (transcript NM_001350162.2) at coding-DNA position 1261, where G is replaced by A; at the protein level this means replaces glycine at residue 421 with serine — a missense variant. Submitter rationale: The missense variant p.G421S in TEX15 (NM_001350162.2) has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The p.G421S variant is novel (not in any individuals) in gnomAD Exomes and is novel (not in any individuals) in 1000 Genomes. There is a small physicochemical difference between glycine and serine, which is not likely to impact secondary protein structure as these residues share similar properties. In silico tools are contradictory in their predictions (SIFT-Tolerated, Polyphen-Damaging) and the residue is not conserved across species. For these reasons, this variant has been classified as Uncertain Significance.
