Likely pathogenic for Abnormal blistering of the skin; Milia; Generalized hypopigmentation; Jaundice; Recessive dystrophic epidermolysis bullosa — the classification assigned by Neuberg Centre For Genomic Medicine, NCGM to NM_000094.4(COL7A1):c.8569G>T (p.Glu2857Ter), citing ACMG Guidelines, 2015: The stop gained p.E2857* in COL7A1 (NM_000094.4) has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The p.E2857* variant is novel (not in any individuals) in gnomAD Exomes and is novel (not in any individuals) in 1000 Genomes. This variant is predicted to cause loss of normal protein function through protein truncation. Loss of function mutations have been previously reported to be disease causing. For these reasons, this variant has been classified as Likely Pathogenic.
