Submitter rationale: The p.V175I variant (also known as c.523G>A) is located in coding exon 4 of the CDK4 gene. The valine at codon 175 is replaced by isoleucine, an amino acid with highly similar properties. This change occurs in the first base pair of coding exon 4. This variant was identified in a cohort of 681 ancestrally diverse, healthy subjects (Bodian DL et al. PLoS One, 2014 Apr;9:e94554). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive.
