NM_000939.4(POMC):c.34del (p.Leu12fs) was classified as Pathogenic by Genetic Services Laboratory, University of Chicago, citing ACMG Guidelines, 2015: DNA sequence analysis of the POMC gene demonstrated two sequence changes in exon 3. The first is a 25bp insertion, c.20_21ins25, that results in an amino acid frameshift and creates a premature stop codon 119 amino acids downstream of the mutation, p.Ser7Argfs*120. The second sequence change is a single base pair deletion, c.34del, that also results in an amino acid frameshift and creates a premature stop codon 58 amino acids downstream of the mutation, p.Leu12Cys*59. These pathogenic sequence changes are predicted to result in an abnormal transcript, which may be degraded, or may lead to the production of a truncated POMC protein with potentially abnormal function. Both next-generation sequencing and Sanger confirmation analysis confirmed that these sequence changes are in trans configuration.
