Pathogenic — the classification assigned by Genetic Services Laboratory, University of Chicago to NM_000162.5(GCK):c.606_607insACACCGT (p.Val203fs), citing ACMG Guidelines, 2015: DNA sequence analysis of the GCK gene demonstrated a seven base pair insertion in exon 6, c.606_607insACACCGT. This pathogenic sequence change results in an amino acid frameshift and creates a premature stop codon four amino acids downstream of the mutation, p.Val203Thrfs*5. This pathogenic sequence change is predicted to result in an abnormal transcript, which may be degraded, or may lead to the production of a truncated GCK protein with potentially abnormal function. This sequence change is absent from large population databases such as ExAC and gnomAD.
