Likely pathogenic — the classification assigned by Genetic Services Laboratory, University of Chicago to NM_014444.5(TUBGCP4):c.1848+4_1848+5del, citing ACMG Guidelines, 2015: DNA sequence analysis of the TUBGCP4 gene demonstrated a sequence change located near the canonical splice donor site in intron 16, c.1848+4_1848+5del. This sequence change does not appear to have been previously described in patients with TUBGCP4-related disorders and has also not been described as a known benign sequence change in the TUBGCP4 gene. Based on in silico splice prediction programs, this sequence change likely affects normal splicing of the TUBGCP4 gene, however functional studies have not been performed to prove this conclusively.
