Pathogenic — the classification assigned by Genetic Services Laboratory, University of Chicago to NM_000474.4(TWIST1):c.108del (p.Arg37fs), citing ACMG Guidelines, 2015: DNA sequence analysis of the TWIST1 gene demonstrated a single base pair deletion in exon 1, c.108del. This pathogenic sequence change results in an amino acid frameshift and creates a premature stop codon 88 amino acids downstream of the mutation, p.Arg37Alafs*88. This pathogenic sequence change is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. This particular pathogenic sequence change has been previously described in a patient with Saethre-Chotzen syndrome (PMID: 16251895).
