NM_021922.3(FANCE):c.952G>A (p.Glu318Lys) was classified as Uncertain significance by Genetic Services Laboratory, University of Chicago, citing ACMG Guidelines, 2015: DNA sequence analysis of the FANCE gene demonstrated a sequence change, c.952G>A, in exon 4 that results in an amino acid change, p.Glu318Lys. This sequence change has been described in the gnomAD database with a frequency of 0.012% in the Latino/admixed American subpopulation (dbSNP rs754914337). The p.Glu318Lys change affects a moderately conserved amino acid residue located in a domain of the FANCE protein that is known to be functional. In-silico pathogenicity prediction tools (SIFT, PolyPhen2, Align GVGD, REVEL) provide contradictory results for the p.Glu318Lys substitution. This sequence change does not appear to have been previously described in individuals with FANCE-related disorders. Due to insufficient evidences and the lack of functional studies, the clinical significance of the p.Glu318Lys change remains unknown at this time.
