NM_000208.4(INSR):c.1001C>T (p.Pro334Leu) was classified as Uncertain significance by Genetic Services Laboratory, University of Chicago, citing ACMG Guidelines, 2015: This sequence change does not appear to have been previously described in patients with INSR-related disorders and has been described in the gnomAD database with a low population frequency of 0.00040% (dbSNP rs373995681). The p.Pro334Leu change affects a moderately conserved amino acid residue located in a domain of the INSR protein that is known to be functional. The p.Pro334Leu substitution appears to be benign using several in-silico pathogenicity prediction tools (SIFT, PolyPhen2, Align GVGD, REVEL). Due to these contrasting evidences and the lack of functional studies, the clinical significance of the p.Pro334Leu change remains unknown at this time.
