NM_000059.4(BRCA2):c.6112C>T (p.His2038Tyr) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H2038Y variant (also known as c.6112C>T), located in coding exon 10 of the BRCA2 gene, results from a C to T substitution at nucleotide position 6112. The histidine at codon 2038 is replaced by tyrosine, an amino acid with similar properties. This alteration has been reported in a cohort of 1010 unrelated patients and families from across India with an indication of breast and/or ovarian cancers (Singh J et al. Breast Cancer Res. Treat., 2018 Jul;170:189-196). This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
