This variant is located within the HNF1A/HNF1B binding region (c.-170 to c.-173 and c.-178 to c.-181) of the P2 promoter of HNF4A, which is defined as critical for the protein’s function by the ClinGen MDEP (PM1). This variant is absent from gnomAD v2.1.1 and v4.1.0 (PM2_Supporting). This variant was identified in an individual with diabetes; however, the calculated MODY probability is <50% (internal lab contributors).
