This variant lies in the ATM gene (transcript NM_000051.4) at coding-DNA position 400, where G is replaced by A; at the protein level this means replaces glycine at residue 134 with serine — a missense variant. Submitter rationale: Variant summary: ATM c.400G>A (p.Gly134Ser) results in a non-conservative amino acid change located in the telomere-length maintenance and DNA damage repair domain (IPR0216680) of the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 1.6e-05 in 251284 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.400G>A in individuals affected with Breast Cancer and no experimental evidence demonstrating its impact on protein function have been reported.
