This variant lies in the RBBP6 gene (transcript NM_006910.5) at coding-DNA position 1783, where G is replaced by A; at the protein level this means replaces alanine at residue 595 with threonine — a missense variant. Submitter rationale: DNA sequence analysis of the RBBP6 gene demonstrated a sequence change, c.1783G>A, in exon 15 that results in an amino acid change, p.Ala595Thr. This sequence change has been described in the gnomAD database with a frequency of 0.047% in the non-Finnish European subpopulation (dbSNP rs199538396). The p.Ala595Thr change affects a highly conserved amino acid residue located in a domain of the RBBP6 protein that is not known to be functional. In-silico pathogenicity prediction tools (SIFT, PolyPhen2, Align GVGD, REVEL) provide contradictory results for the p.Ala595Thr substitution. This sequence change does not appear to have been previously described in individuals with RBBP6-related disorders. Due to insufficient evidences and the lack of functional studies, the clinical significance of the p.Ala595Thr change remains unknown at this time.
