Pathogenic — the classification assigned by Genetic Services Laboratory, University of Chicago to NM_001197104.2(KMT2A):c.2569G>T (p.Glu857Ter), citing ACMG Guidelines, 2015: DNA sequence analysis of the KMT2A gene demonstrated a de novo sequence change, c.2569G>T, which results in the creation of a premature stop codon at amino acid position 857, p.Glu857*. This sequence change is absent from population databases such as ExAC and gnomAD. This pathogenic sequence change is predicted to result in an abnormal transcript, which may be degraded, or may lead to the production of a truncated KMT2A protein with potentially abnormal function.
