Likely pathogenic — the classification assigned by Genetic Services Laboratory, University of Chicago to NM_015346.4(ZFYVE26):c.6384C>A (p.Tyr2128Ter), citing ACMG Guidelines, 2015: DNA sequence analysis of the ZFYVE26 gene demonstrated a sequence change, c.6384C>A, which results in the creation of a premature stop codon at amino acid position 2128, p.Tyr2128*. This sequence change is predicted to result in an abnormal transcript, which may be degraded, or may lead to the production of a truncated protein with potentially abnormal function. This sequence change has not been previously described in patients with spastic paraplegia 15 and has also not been described as a known benign sequence change in the ZFYVE26 gene.
