Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant was absent in 250990 control chromosomes. c.458C>T has been observed in a homozygous individual affected with D-Bifunctional Protein Deficiency (Ferdinandusse_2006). These data indicate that the variant may be associated with disease. At least one publication reports experimental evidence evaluating an impact on protein function, however, the variant protein could not be studied with respect to its enzymatic properties as it rapidly degraded after purification following expression in E.coli (Mehtala_2013).
