Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001025295.3(IFITM5):c.143A>G (p.Asn48Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces asparagine, which is neutral and polar, with serine, which is neutral and polar, at codon 48 of the IFITM5 protein (p.Asn48Ser). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individuals with clinical features of IFITM5-related conditions (PMID: 34156493). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 1333457). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
