NM_007055.4(POLR3A):c.1910-1G>A was classified as Likely pathogenic for Ankle clonus; Global developmental delay; Hyperreflexia; Leg dystonia; Developmental regression; Spastic diplegia; Leukodystrophy, hypomyelinating, 7, with or without oligodontia and/or hypogonadotropic hypogonadism by 3billion, citing ACMG Guidelines, 2015. This variant lies in the POLR3A gene (transcript NM_007055.4) at the canonical splice acceptor site of the intron immediately before coding-DNA position 1910, where G is replaced by A; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: Canonical splice site: predicted to alter splicing and result in a loss or disruption of normal protein function through protein truncation. Multiple pathogenic variants are reported in the predicted truncated region (PVS1_VS). It is not observed in the gnomAD v2.1.1 dataset (PM2_M). Therefore, this variant is classified as likely pathogenic according to the recommendation of ACMG/AMP guideline.
