This variant lies in the PRKAG2 gene (transcript NM_016203.4) at coding-DNA position 469 through coding-DNA position 471, duplicating 3 bases; at the protein level this means duplicates serine at residue 157. Submitter rationale: This variant causes an in-frame duplication of one amino acid at exon 4 of the PRKAG2 protein.To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with PRKAG2-related disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
