Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_002474.3(MYH11):c.3030C>G (p.Asp1010Glu), citing Ambry Variant Classification Scheme 2023: The p.D1010E variant (also known as c.3030C>G), located in coding exon 23 of the MYH11 gene, results from a C to G substitution at nucleotide position 3030. The aspartic acid at codon 1010 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
