NM_001369.3(DNAH5):c.4924G>A (p.Gly1642Arg) was classified as Uncertain significance for Primary ciliary dyskinesia 3 by Johns Hopkins Genomics, Johns Hopkins University, citing ACMG Guidelines, 2015: This DNAH5 variant (rs150050027) has been identified in a large population dataset and the minor allele frequency is neither low enough to consider the variant rare (<0.1%) nor high enough to consider it a population polymorphism (>1%) within the African/African American subpopulation (gnomAD: 36/24968 alleles; 0.1442%, no homozygotes). It has has not been reported in ClinVar nor the literature, to our knowledge. Three bioinformatic tools queried predict that this substitution would be damaging and the glycine residue at this position is evolutionarily conserved across all species assessed except American alligator. We consider the clinical significance of DNAH5 c.4924G>A to be uncertain at this time.
