Likely pathogenic for Primary ciliary dyskinesia 2 — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001256715.2(DNAAF3):c.548_558dup (p.Ser187delinsArgSerProTer), citing LabCorp Variant Classification Summary - May 2015: Variant summary: C19orf51 (DNAAF3) c.752_762dup11 (p.Ser255ArgfsX4) results in a premature termination codon, predicted to cause a truncation of the encoded protein or absence of the protein due to nonsense mediated decay, which are commonly known mechanisms for disease. Truncations downstream of this position have been cited in ClinVar and HGMD as pathogenic and disease-associated. The variant was absent in 131332 control chromosomes (gnomAD). To our knowledge, no occurrence of c.752_762dup11 in individuals affected with Ciliary Dyskinesia, Primary, 2 and no experimental evidence demonstrating its impact on protein function have been reported. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as likely pathogenic.
