NM_000540.3(RYR1):c.11969G>T (p.Gly3990Val) was classified as Pathogenic for Malignant hyperthermia of anesthesia by Laboratory for Molecular Medicine, Mass General Brigham Personalized Medicine, citing ACMG Guidelines, 2015: The p.Gly3990Val variant in RYR1 has been reported in at least 14 European individuals with malignant hyperthermia and segregated with disease in at least 7 affected individuals (Carpenter 2009, Merritt 2017, Miller 2018). It is absent from gnomAD (http://gnomad.broadinstitute.org). This variant has also been reported in ClinVar (Variation ID 133027) and was classified as drug response allele on 6/14/2019 by the PharmGKB ClinGen-approved expert panel. In vitro functional studies provide some evidence that this variant impacts protein function (Merritt 2017).
