Uncertain Significance for Malignant hyperthermia, susceptibility to, 1 — the classification assigned by All of Us Research Program, National Institutes of Health to NM_000540.3(RYR1):c.11953T>C (p.Trp3985Arg), citing ACMG Guidelines, 2015. This variant lies in the RYR1 gene (transcript NM_000540.3) at coding-DNA position 11953, where T is replaced by C; at the protein level this means replaces tryptophan at residue 3985 with arginine — a missense variant. Submitter rationale: This missense variant replaces tryptophan with arginine at codon 3985 of the RYR1 protein. Computational prediction suggests that this variant may have deleterious impact on protein structure and function. To our knowledge, functional studies have not been reported for this variant. This variant has been reported in three individuals affected with malignant hyperthermia susceptibility, including two related individuals with a family history of a clinical malignant hyperthermia reaction (PMID: 18719443, 23558838). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
