NM_001365276.2(TNXB):c.8323G>A (p.Gly2775Arg) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G2775R variant (also known as c.8323G>A), located in coding exon 23 of the TNXB gene, results from a G to A substitution at nucleotide position 8323. The glycine at codon 2775 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
